MOTS-c is a mitochondrial-derived peptide — a short signalling molecule encoded not in the cell’s nuclear DNA but within the genome of the mitochondria themselves. This unusual origin is part of what has made it a subject of scientific interest, as it represents a form of communication running from the mitochondria outward to the rest of the cell.

Much of the research attention given to MOTS-c stems from its apparent role in cellular energy metabolism. Studies have examined its influence on how cells process glucose and respond to insulin, along with its connection to the activation of AMPK — a pathway often described as a master regulator of energy balance, and one the body also engages during physical activity. It is this overlap that has led some researchers to investigate MOTS-c as a possible “exercise mimetic,” a compound that may reproduce certain metabolic signals normally associated with exercise. Much of this work has been conducted in animal models and remains preliminary.

Further research has explored its potential relationship to physical endurance, metabolic stress resistance, and the decline in mitochondrial efficiency that accompanies ageing — making it a recurring subject of laboratory investigation into how mitochondrial signalling shapes energy homeostasis across the body.
